3. Reaction Principle
S-2238 is a specific substrate for thrombin, with a structure designed to mimic the N-terminal region of the alpha chain of fibrinogen. Chemically modified to enhance stability and reactivity, it plays a critical role in detection. During testing, excess thrombin is added to the plasma sample, where antithrombin forms a 1:1 complex with thrombin. The remaining thrombin reacts with the chromogenic group p-nitroaniline of S-2238, producing coloration. The intensity of the color correlates positively with residual thrombin and negatively with antithrombin. By measuring absorbance changes through spectrophotometry, the activity can be precisely determined.
