I. Working Principle
U-3ʹ-BCIP is a high-sensitivity chromogenic substrate specifically designed for detecting the activity of Ribonuclease A (RNase A). Its mechanism is based on enzyme-specific hydrolysis: under the specific catalysis of RNase A, the phosphodiester bond in the substrate molecule is hydrolyzed, releasing an indole group. This group undergoes oxidative dimerization to form an insoluble blue/purple precipitate, accurately localizing the enzyme's active site.
The value of this mechanism, based on enzymatic reaction and colorimetric output, extends far beyond theory:
High Signal-to-Noise Ratio: The precipitating signal does not easily diffuse, resulting in extremely low background.
